Can an Ambush unit get Mask of the Brooding bonus after Akali's trigger?

No. The Ambush unit is designated defender in the first cleanup after it resolves, before Akali's ability resolves. At that time it is not alone, so the Mask does not proc. There is no timing to get the Mask bonus with Ambush here. Grim Apothecary has the same issue.
